(a,b) HEK cells were cotransfected with Rluc-PTEN-YFP and TPαR or M1MR, and treated with thromboxane mimetic U46619 (1 μM) or Carbachol (CCh) (100 μM). Graphs represent the means±s.e.m. of three independent experiments. ***P<0.001, *P<0.05, t-tests. (c,d) HeLa cells were cotransfected with TPαR or M1MR, and control or PTEN-targeting siRNA. Cells were left unstimulated or stimulated with U46619 (1 μM) or Carbachol (CCh) (100 μM) before being lysed and assessed for pAKT and PTEN levels by western blotting. BRET measurements in these experiments were performed on adherent cells.
