In clockwise order, 1 represents the catalytic channel undergoing conformational transitions, 2 indicates how substrate binding allosterically activates the enzyme, 3 indicates catalysis, and 4 and 5 indicate regeneration of the enzyme. The colour changes of a Ubl-specific protease represent allosteric effects due to binding of the β-grasp domain.
