(a) AND-Tg T cells expressing a mock-monomeric EGFP or WT, V801P or L815P CARMA1-mEGFP (green) were stained for TCRβ with DyLight 649-conjugated H57 F′ab (red), plated onto an MCC88-103-prepulsed planar bilayer containing I-Ek-, ICAM-1- and CD80-GPI and imaged by confocal microscopy at 2 and 30 min after contact. Images of cells before attachment to the bilayer were taken at the equatorial plane of the cells. The percentage of cells forming CARMA1 microclusters at 30 min after contact is shown in b (n=50). A representative of three independent experiments is shown. Scale bars, 5 μm.
