(a) Effects of L-cysteine concentration on the amount of L-alanine produced by the NIUF complex. The reactions were performed with the NIUF complex (20 μM) and increasing amounts of L-cysteine in the absence of DTT. L-alanine production was monitored by ADH assays after 10 min of reaction. (b,c) Effect of FXN and ISCU on the initial velocities of L-alanine formation under steady-state conditions. The NI complex (20 μM) alone and with equimolar amounts of FXN (b) and the NIU and NIUF complexes (20 μM) (c) were incubated with various amounts of L-cysteine and the initial velocities were plotted as a function of L-cysteine concentrations. The bars represent the s.e. from three independent experiments. The lines represent the linear regression.
