Figure 5

(a,b) Mab21-l3 MO decreases α-tubulin expression. The indicated MO (30 ng each) was co-injected with fluorescein dextran (100 ng) into the left ventral blastomere at the four-cell stage, and embryos at stage 13 (a) or 17 (b) were subjected to whole-mount in situ hybridization against α-tubulin. Ventral (a) or lateral (b) views of embryos are shown in left 6 (a) or upper 12 (b) panels. A, anterior; P, posterior in b. α-tubulin expression was decreased in mab21-l3 MO-injected embryos (stage 13, 100%, n=15; stage 17, 100%, n=15), but not affected in those injected with control MO (stage 13, 100%, n=14; stage 17, 100%, n=14) or mab21-l3 5mis MO (stage 13, 100%, n=6; stage 17, 88%, n=8). The middle (a) or lower (b) 12 panels show higher magnification of boxed areas in left 6 (a) or upper 12 (b) panels. Scale bars, 500 μm (a, left panels; b, upper panels) and 50 μm (a, middle panels; b, lower panels). The right graphs show the ratio of the α-tubulin-expressing cell number per field in the MO-injected side to that in the uninjected side. Shown is the average from embryos in one (mab21-l3 5mis MO) or two (control MO and mab21-l3 MO) independent experiments. n, the number of embryos scored. The error bar represents s.d. **P<0.01 by Tukey’s test.
