Time-dependent 20S proteasome degradation assays of α-synuclein and the C-terminal region of p53 (Cp53, residues 292–393), in the presence or absence of DJ-1WT or DJ-1C106A. At the indicated time points, aliquots were quenched and evaluated by SDS–PAGE (top panel), followed by chemiluminescence quantification (bottom panel). In contrast to DJ-1WT, DJ-1C106A has a notably reduced capacity to rescue α-synuclein and Cp53 from degradation. Error bars represent the s.d. of three repeats.
