(a–c) Quantitative RT–PCR analysis of ERK7 expression levels. The average of three independent experiments is presented. The expression levels of ERK7 were normalized to those of ODC. (a) Each mRNA (500 pg per cell) was injected into the animal region of four blastomeres at the four-cell stage. Injected embryos were harvested at stage 13 and assayed for ERK7 mRNA by quantitative RT–PCR analysis. (b) Luciferase mRNA (750 pg per cell) alone, NICD mRNA (250 pg per cell) plus Luciferase mRNA (500 pg per cell), or NICD mRNA (250 pg per cell) plus FoxJ1 mRNA (500 pg per cell) was injected into the animal region of the four blastomeres at the four-cell stage. Animal caps were isolated at stage 10. The animal caps were harvested at the equivalent of stage 19–20 and assayed for ERK7 mRNA by quantitative RT–PCR analysis. (c) Each MO (20 ng per cell) was injected into the four blastomeres at the four-cell stage. Animal caps were isolated at stage 10.
