Sequential ChIP–PCR anti-SUZ12 or anti-BMI1 and then anti-SALL4 were performed from chromatin of control ZHBTc6 (+tetracyclin) ESCs (CTRL) or OCT4-overexpressing (ZHBTc6−tetracyclin) mesendodermal cells (Oct4OE). Both Sox-2C (a) and Sox-17 (b) enhancers and promoters were interrogated. Sox2B and Sox17R2 regions were used as negative controls.
