Dnmt1Δ/Δ mice were crossed with MMTV-Neu-Tg and C3(1)-SV40-Tg mice to generate MMTV-Neu and C3(1)-SV40 transgenic mice in wild-type and Dnmt1-null genetic backgrounds. Using these mice, we monitored (a) tumour incidence, (b) age of the animals at tumour onset, (c) tumour size, (d) lung metastasis and (e,f) average survival time. Data are mean±s.e.m. (n=24 mice in each group). (g,h) Representative contour blots show reduced CSC (CD49f+CD24+) in Dnmt1Δ/Δ-MMTV-Neu and Dnmt1Δ/Δ-C3(1)-SV40 mice compared with their respective controls. Bar diagrams represent per cent CSC populations of respective mouse mammary tumours. Data are mean±s.e.m. (n=5 mice in each group). Statistical analysis was performed using unpaired Student’s t-tests. Error bars represent s.e.m. of independent experiments.
