Whole-cell lysates were prepared from TH1 and TH2 cultures and incubated with either an isotype Ig control antibody (isotype), a pan hnRNP antibody (recognizes a conserved hnRNP amino acid sequence) or a WDR5 antibody. Immunoprecipitates were harvested with protein A/G sepharose beads, RNA was isolated and the lncRNA transcripts in the immunoprecipitates were determined by quantitative PCR. Fold enrichment±s.d. (n=3) was determined by comparing the specific Ct obtained from the isotype immunoprecipitate to the antibody-specific immunoprecipitate by calculating 2(IsotypeCt-Ab-specificCt). *=P<0.05 determined using Student’s t-test.
