(a) Spot test. 10⁶, 10⁵, 10³ and 10² cells with the pGAL-NDD1 plasmid or empty vector were spotted on raffinose and galactose SD uracil-deficient plates and grown for 2 days at 30 °C. (b) Morphology of cells expressing gal-inducible NDD1, Cdh1^m11 and both NDD1 and Cdh1^m11. Cells were grown to mid-log phase in raffinose, supplemented with galactose and sampled at the indicated time points. (c) The fraction of normal cells, cells with elongated buds and large cells was quantified in duplicates of three independent experiments, n=200 each (1200 in total). Error bars represent s.d. of these three experiments. The length of the scale bar is 10 μm. (d) The indicated strains were subjected to the same experimental conditions as in b and sampled for immunoblotting at the indicated time points.
