Figure 1: AF6 is expressed at low levels in human pancreatic cancer.
From: Loss of polarity protein AF6 promotes pancreatic cancer metastasis by inducing Snail expression

(a) Western blots of protein extracts derived from immortalized human pancreatic ductal epithelial cell line HPDE and nine other human pancreatic cancer cell lines; l-afadin- or s-afadin-overexpressing Capan-1 cells were used as positive controls. The anti-AF6 antibody recognizes an epitope corresponding to the last C-terminal amino-acid residues of l-afadin. The anti-AF6 antibody recognizes an epitope corresponding to the PDZ domain, which is present in both l-afadin and s-afadin. Up and down blank arrows indicate l-afadin or s-afadin, respectively. (b) Real-time PCR analysis of the mRNA expression of AF6 in 40 paired human pancreatic cancer tissues (T) and adjacent normal tissues (N). Data represent the mean±s.d. (n=3). (c) Box plots comparing AF6 mRNA levels in normal pancreas and pancreatic cancer in published data sets from Oncomine. **P<0.01, ***P<0.001, Student’s t-test. (d) The mRNA expression profiles of AF6 in normal pancreas, different stages of PanIN, and in PDAC. *P<0.05, **P<0.01, Student’s t-test. (e,f) IHC staining for AF6 in adjacent normal and paired pancreatic cancer tissues (e). Box plots of AF6 protein expression assessed by blinded IHC analyses of 90 normal and paired pancreatic cancer tissues (f). ***P<0.001, Student’s t-test. Scale bar, 100 μm.
