4 μM (a) Rab1Q67L, L125C:GTP, (b) Rab1Q67L, L125C–aGTP, (c) Rab1E35C, Q67L:GTP and (d) Rab1E35C, Q67L–aGTP were incubated with 0.4 μM TBC1D2014-305. The resulting progress curves (black) were fitted using a single exponential equation (red line) and yielded observed rate constants of 6.0 × 10−4 s−1 (Rab1Q67L, L125C:GTP), 5.0 × 10−4 s−1 (Rab1Q67L, L125C–aGTP), 2.0 × 10−4 s−1 (Rab1E35C, Q67L:GTP) and 1.1 × 10−4 s−1 (Rab1E35C, Q67L–aGTP).
