(a) RAW264.7 cells were transduced with lentivirus encoding constitutively active Notch1 ICD expressed from bicistronic IRES–GFP or IRES–GFP alone. Zymosan-induced oxidative burst was measured by luminol chemiluminescence as in Fig. 2a–c. Primary bone marrow-derived macrophages were transduced with lentivirus encoding the indicated shRNAs. After 6 h of treatment with the indicated stimuli, macrophages were lysed and subjected to transcriptional profiling by Fluidigm (b) and qPCR (c). Data represent the mean of triplicates±s.d. *P≤0.05 as determined by Student’s t-test.
