(a,b) The HAT assay and downstream DNA and data processing was performed using DNL-1 identically as described in Fig. 3b with an α-H3K18ac specific antibody in the absence (blue) or presence (gray) of 20 mM Nω-Acetylhistamine (NAH, a) or in the absence (blue, b) or presence of 20 μM Bio-H4Kac4 (green, b) or 100 µM Bio-H4Kac4 (dark gray, b). For (b), the biotinylated H4 peptide was removed prior to α-H3K18ac affinity pulldown using streptavidin-coated magnetic beads. Internal normalization: H3Kac5 variant, set as 1 (red asterisks). Values are shown as (mean ± SD, n=3) or plotted independently (n=2) employing the same grid as in Fig. 1b.
