Type I interferons (IFNs) produced by pDCs had been previously implicated in the early phase of diabetes in humans and non-obese diabetic (NOD) mice. In accordance with this, the authors found that IFNα expression peaks at 3 to 4 weeks of age and precedes pro-inflammatory gene expression in NOD mice but not in control mice. High IFNα levels coincided with high pDC numbers in the islets of NOD mice and, consistent with previous reports, antibody-mediated pDC depletion delayed the development of diabetes. Furthermore, the authors found that pDCs produced IFNα and promoted diabetogenic T cell responses following Toll-like receptor 7 (TLR7) or TLR9 stimulation.
