OSM induces EMT in breast cancer cells and enhances cell migration and invasion. (a) MCF-7 cells were treated with 10 ng/ml OSM and the expression levels of E-cadherin, fibronectin, vimentin, ZEB1 and Snail were analyzed by western blot at the indicated time points. (b) MCF-7 cells were treated with phosphate-buffered saline or 10 ng/ml of OSM for 5 days. The expression levels of E-cadherin and fibronectin were examined by immunofluorescence/confocal microscopy. Bar=50 μM. (c) The migratory activities were determined by the wound-healing assays at the indicated days after OSM stimulation. (d, e) The invasive activities of MCF-7 cells were determined by Matrigel invasion assays. The invading cells were stained (d) and quantified (e). Error bars represent s.e. (N⩾3).
