Impact of therapeutics on EGF-induced fast signaling. Data show EGF-induced ERK1/2 (a and c) and AKT (b and d) phosphorylation after preincubation of SKBR3 cells with trastuzumab, pertuzumab or erlotinib (a and b), as well as with all possible combinations (c and d). Phosphorylation dynamics were analyzed by RPPA. EGF (5 nM) induced phosphorylation of AKT and ERK1/2 in serum-starved SKBR3 cells. Pertuzumab and trastuzumab downregulated AKT signaling, whereas erlotinib, applied alone and in all combinations with therapeutic antibodies, inhibited the activation of both the pathways. Corresponding data set for HCC1954 cells is shown as Supplementary Figure S3 and Supplementary Table S1.
