Depletion of OLFM4 induces NF-κB activation and IL-8 expression in gastric cancer. (a) HGC-27 and AGS cells were transfected as indicated. Forty-eight hours later, nuclear and cytoplasmic proteins were extracted and subjected to western blot analysis. (b) The effect of OLFM4 siRNA on IL-8 mRNA levels in HGC-27 and AGS cells were detected by real-time PCR. (c) The effect of OLFM4 siRNA on IL-8 protein levels in the supernatant of HGC-27 and AGS cells were detected by ELISA. (d) The effects of Reparixin (100 nM) on the migration of OLFM4-depleted gastric cancer cells were detected by transwell assay. (e, f) Correlation of OLFM4 with the expression of IL-8 in clinical EGC cases. Representative images in consecutive tumor sections were shown. Correlation between OLFM4 and IL-8 protein expression in 73 cases was analyzed.
