Abstract
β-Adrenergic receptor (βAR) has been previously demonstrated in human placental membranes. Its demonstration in placentas of multiple species and trophoblastic tumors might support its regulatory function on the trophoblastic cell itself. βAR in rat, guinea pig, rabbit and human placenta and trophoblastic tumors were therefore examined. Term placentas were obtained, dissected to remove decidua and processed in identical fashion. The β-adrenergic antagonist dihydroalprenolol, (−)[3H]DHA bound to a single, saturable class of sites in all placental membranes. Placental βAR was coupled with catecholamine sensitive adenylate cyclase in all species.
βAR was absent in two human choriocarcinoma cell lines (JEG-3, BeWo) but was present in a hydatidiform mole obtained freshly from surgery (91 fmoles/mg, KD=1.31 nM). We conclude that 1) variable but consistently high density βAR is present coupled to adenylate cyclase in mammalian placenta; 2) the affinity (KD) of the βAR is similar in all species; and 3) the presence of high density βAR on a hydatidiform mole containing almost totally trophoblast tissue suggests a βAR is located on trophoblastic cell membranes.
