The rapid neonatal bone mineral accretion rate is adversely affect by factors that alter systemic pH and plasma mineral concentrations. Examples include respiratory and cardiovascular diseases, diuretic therapies, and nutritional status. In an effort to quantitate the effects of alterations in local pH and concentrations of inorganic phosphate, ionized calcium (Ca++), and ionized magnesium (Mg++) on neonatal bone, a bone disc bioassay was utilized, with bone discs harvested from the calvaria of 21 day-old rats. For each measurement, 6-9 bone discs were incubated in physiologic medium (isotonic saline plus Ca++ 1.25 mM + Mg++ 0.55 mM + phosphate 6.0 mg/dl, pH 7.39), or similar test media changed by modifying the concentration of one physiologic parameter at a time. Bone disc uptake of Ca++ was measured, and results expressed as the percent difference (increase or decrease) in Ca++ uptake of the test media compared to physiologic.
