Effect of Allopurinol (ALLO) and Deferoxamine (DFO) on Cerebral Blood Volume (CBV) and Cerebral Tissue Oxygenation (Δ Cytaa3) after Hypoxic-Ischemic (HI) Reperfusion Injury in Newborn Piglets

Free radical-induced postasphyxial reperfusion injury has been recognized as an important cause for neonatal brain damage. We investigated if post-HI treatment with ALLO (xanthine oxidase inhibitor) or DFO (free iron chelator) preserved post-HI CBV (as proportional to tHb) and Cytaa3, using near infrared spectroscopy. Severe HI was induced in 12 newborn piglets (1-3 days) after occluding the common carotid arteries for 1 h combined with a reduced FiO2 (0.08-0.10). Temperature was maintained at 38-39 °C. Following HI 4 piglets received ALLO (20 mg/kg/i.v.), 4 received DFO (10 mg/kg/i.v.) and 4 received a placebo (PLAC). CBV and ΔCytaa3 were measured for 30 minutes pre- and at 3, 6 and 24 h post-HI.
