Two regions of homology (blue rectangles) are shown on either side of a DSB. Step 1: a 5′ > 3′ resection occurs at both ends, exposing the homology. Step 2: in a RAD52-dependent fashion, the single-stranded ends are annealed together. Step 3: residual 3′-flaps (red lines) are nucleolytically trimmed away. Step 4: the gaps are filled in with polymerases, and any nicks are ligated to complete the repair process.
