B12 impacts on IL-7/IL-7R-mediated signaling and cell viability. a IL-7 starved TAIL7 cells were incubated with 125 nm (18 μg/mL) of B12 and 50 ng/ml of IL-7 together for 120 min at 37 °C. Signaling pathway activation was analyzed by immunoblot using the indicated phospho-specific antibodies (left). Viability was assessed at 120 h (right). b Serum starved HPB-ALL cells were treated in a similar manner. Effects on viability were recorded at 120 h. c IL-7R mutant DND4.1 cells and d D1 cells stably transduced with mutant IL-7Rα (D1mutP2) were treated with B12 and cell survival was analyzed at 120 h. Statistical analysis was performed using two-tailed unpaired t test with Welch´s correction (**** p < 0.0001, *** p < 0.001, ** p < 0.01, * p < 0.05). Values indicate the mean and SEM of three replicates. In all experiments an irrelevant IgG isotype was used as negative control
