Schematic diagram of CRYAB on the activation of the NF-κB pathway by TNF-α. On TNF-α stimulation, the TNF-α receptor is activated, followed by the activation of TAK1, which is the crucial pivot for the important intersection of the NF-κB pathway activation. Thereafter, the activation of the IKK complex and subsequent degradation of IκBα via the phosphorylation catalytic subunit IKKβ are activated. According to the aforementioned results, we found that CRYAB does not affect the degradation of IKK complexes, but inhibits IKKβ activity via binding to IKKβ and IKKα and interfering IKK complex formation
