Effect of miR-219a-5p on Th1/Th17 cell-related gene expression. PB-CD4+ T cells isolated from healthy controls (HC, n= 6), CD (CD, n = 4) and UC (UC, n = 4) patients were activated with plate-bound anti-human CD3 (5 μg/ml) and anti-human CD28 (2 μg/ml) mAs for 48 h, and then transduced with LV-miR-219a-5p, LV-miR-219a-5p sponge and LV-control for 6 h, respectively. These cells were then cultured in medium for an additional 5 days. Total RNA was isolated from these cells and mRNA levels of IFN-γ (a), IL-17A (b), TNF-α (c), IL-21 (d), T-bet (e) and RORC (f) were detected by qRT-PCR. *p < 0.05, **p < 0.01 versus LV-control transduced CD4+ T cells.
