SARS-CoV-2 N protein dually regulates viral replication. a, c Caco-2 cells were infected with SARS-CoV-2 at MOI of 0.01. At 6, 12, 24, and 48 hpi, total RNA was extracted. The mRNA expression of the viral N gene (a) and host IFNA, IFNB1, ISG15 (c) relative to GAPDH control were examined using qPCR. b Caco-2 cells were infected with SARS-CoV-2 as indicated as in a, HEK293T cells were transfected with the indicated 2N plasmids. At 24 hpt, and lysis of infected and transfected cells (50 μg/sample) were analyzed by immunoblot. d–f The HEK293T cells were transfected with 2N plasmids as indicated.
