Schematic overview of competition binding assay. In this assay, DNA-tagged kinases are incubated with immobilized probes that bind kinases at the ATP-binding pocket. Then, a free test compound is introduced to compete with the probe to bind its target kinase, and the selectivity of a test compound is evaluated by quantifying the DNA-tagged kinase associated with the solid bead by real-time quantitative PCR
