From: ERK/mTOR signaling may underlying the antidepressant actions of rapastinel in mice

Our results showed ERK-mediated mTOR signaling may play critical in the antidepressant-like effects of repeated treatment with rapastinel in mice. Our data further indicated that these effects may be mediated partly through the ability of rapastinel to enhance VGF and BDNF expression and then the VGF/BDNF/TrkB autoregulatory feedback loop by increasing the ERK/mTOR pathway. The VGF/BDNF/TrkB autoregulatory feedback loop may supply the VGF and BDNF to facilitate the AMPAR-mediated fast releases in the rapid-acting antidepressant actions of single or repeated treatment with rapastinel.
