Well-established and validated method. Relatively fast and accessible.

qPCR is an indirect method that requires the use of standards for quantification. It can be subject to variations depending on amplification efficiency and biases introduced during analysis.

ddPCR

Absolute quantification of VCN, eliminating the need for a standard curve. High precision, sensitivity, and reproducibility. Less prone to variations than qPCR, allowing detection of copy number variations even in heterogeneous samples.

Expensive method requiring specialized equipment. May be slower than qPCR in some laboratories.
