Numb and Numblin osteoblasts maintain the physiological bone mass. a When expressed normally in osteoblasts, the NUMB-NEDD4-PTEN tri-complexes produce, leading to ubiquitin-mediated proteasomal degradation of PTEN. At the same time, GLI1 tends to be degraded in proteasomes. Akt and Hedgehog signals are moderate in osteoblasts. b When Numb is suppressed, PTEN and GLI1 accumulate in the cytoplasm where they inhibit Akt to suppress bone formation and activate Hedgehog to enhance bone resorption, respectively
