ZH-161 or ZH-305 cells were plated at 300 cells/well in NB medium in the absence or presence of SD-208 (1 µM) or TGF-β2 (10 ng/ml) or SD-208 combined with TGF-β2 in 96-well plates. c-MET shRNA expressing or EMD1214063 (200 nM)-treated ZH-161 or ZH-305. Sphere formation was assessed at day 21 in triplicates (*p < 0.05, effects of TGF-β2 compared to control, # p < 0.05, effect of TGF-β2 and SD-208 co-treatment compared to TGF-β2 alone; ω p < 0.05, effect of EMD1214063 compared to control, θ p < 0.05, effect of c-MET shRNA compared to control). Spherogenicity was calculated based on the number of spheres and average size of the spheres as indicated (upper panels).
