U937 cells were pretreated with 20 μM of Y27632, a ROCK1 inhibitor, for 2 h, followed by treating with 10 μM of MC-3129 for 24 h. a, b Total cellular extracts, cytosolic (C) and mitochondrial (M) fractions were prepared and subjected to western blot analysis using antibodies against ROCK1, p-PTEN, PTEN, p-Akt, Akt, p-cofilin, cofilin, GADPH, and Cox IV. c, d Apoptosis was determined by flow cytometry with Annexin V/PI staining and total protein lysates were analyzed by immunoblotting using the indicated antibodies.
