U937 cells were stably transfected with lentivirus containing ROCK1-specific siRNA (shROCK1) or a scrambled control siRNA (shCon), cells were treated with or without 10 μM of MC-3129 for 24 h. a, b Total cellular extracts, cytosolic (C) and mitochondrial (M) fractions were prepared and subjected to western blot analysis using anti-ROCK1, p-PTEN, PTEN, p-Akt, Akt, p-cofilin, cofilin, GADPH, and Cox IV. c, d Apoptosis was measured by flow cytometry using Annexin V/PI staining, and total protein lysates were analyzed by immunoblotting using the indicated antibodies.
