ASCs were primed with 0.5 μg/ml LPS for 4 h, prior to challenging with 1 μg/ml LPS, 0.25 μg/ml flagellin, 2 μg/ml dsDNA, and 10 μM nigericin respectively, with or without 0.025% v/v FuGENE HD. Pyroptosis was detected by immunoblot analysis (a) of GSDMD-NT and mature IL-1β in ASCs, ELISA detection of mature IL-1β (b) and LDH release (c) in cell supernatant. n = 3. Tubulin was used as an internal control for immunoblot analysis. Data are representative of at least three independent experiments. Statistical significance between the control group and treated groups was determined using one-way analysis of variance. Data are shown as mean ± SD. ns: p > 0.05, *p < 0.05.
