A, B The TOPFlash plasmid was used to examine the effects of ADCK1 on the transcriptional activity of the β-catenin/TCF4 complex. C, D qPCR was performed to examine the effects of ADCK1 on the expression of Axin2. E DN β-catenin abolished the effects of ADCK1 on the migration of RKO cells. The migrated cells were counted, and the expression of ADCK1 and DN β-catenin was confirmed using western blotting. F DN β-catenin abolished the effects of ADCK1 on the anchorage-independent growth of RKO cells. Fourteen days after seeding in the soft agar assay, the colony numbers were determined and analyzed.
