A HEK293T cells were transfected with Flag-GSDMD and Myc-HDAC4, treat with or without TSA. Proteins were immunoprecipitated with anti-acetyl-lys antibody (Ace-lys), and the precipitates were analyzed using an anti-Flag or anti-Myc antibody. B NC and sg-HDAC4 HEK293T cells were transfected with Flag-GSDMD, together with or without Myc-HDAC4. GSDMD acetylation was analyzed by immunoprecipitation with an anti-acetyl-lys antibody followed by western blotting. C HEK293T cells were transfected with Flag-GSDMD and Myc-HDAC4 or HDAC4-803L, treated with or without TSA. GSDMD acetylation was analyzed by immunoprecipitation. LDH release of HEK293T cells transfected with GSDMD-p30 (D) or GSDMD-FL and Caspase-1 (E) or GSDMD-FL and Caspase-4 (F), together with HDAC4-WT or its mutants.
