A Coomassie blue staining of biotinylated SH3PXD2A-AS1-associated proteins is shown. B RNA pulldown assays were performed to verify the mass spectrometry results. Biotin-SH3PXD2A-AS1 and antisense RNA were obtained by in vitro transcription by using T7 RNA polymerase. Western blotting was performed to determine DHX9, HSPA8 and HSPA5. C RIP assays were performed to test the interaction of SH3PXD2A-AS1 and DHX9. Relative quantification of SH3PXD2A-AS1 and 18 S rRNA in RNA-protein complexes immunoprecipitated with IgG or DHX9 from whole cell extracts; 18 S rRNA was used as a negative control binding RNA.
