The listed skin SCC cells, primary human skin fibroblasts (“Fibroblasts”) and keratinocytes (“Keratinocytes”), stably expressing the lentiviral POLRMT shRNA (“shPOLRMT”) or the scramble control shRNA (“sh-C”), were established, POLRMT mRNA expression was tested (A and G). Cells were further cultivated for applied time periods, cell proliferation, migration, ROS production, depolarization of mitochondria and apoptosis were tested by nuclear EdU staining (B), “Transwell” (C), CellROX staining (D), JC-1 staining (E) and caspase-3 activity (H)-TUNEL staining (F and I) assays, respectively, and results were quantified. *P < 0.05 versus “sh-C” cells.
