AV/PI double staining was performed in different MPNST cell lines, including S462 (A), ST8814 (B), STS26T (C), and T265 (D). The MPNST cells were treated with 0-50 μM DDR1-IN-1 as indicated for 24 hours, the AV/PI double staining was manipulated as Materials and Methods described, and the fluorescent intensity was detected through flow cytometry (n = 3). The PI-positive cell (%) was plotted in the bar chart and the p-value was shown. *P < 0.05, **P < 0.01, ***P < 0.001, ****P < 0.0001. E Total protein was extracted after treating ST8814 cells with 0-5 μM DDR1-IN-1 for 24 hours and the necroptosis-related markers were evaluated. F The immunofluorescent images of control and DDR1-IN-1 treated S462 cells are shown. The green fluorescent represents the p-MLKL, and the red arrows indicate the cells undergoing necroptotic cell death.
