YAP/TEADs complex plays a pivotal role in PDAC via transcription of downstream effectors that directly induce cell transformation and proliferation (STAT3, LIFR, BMP4, CTGF, COX2, MMP7, BCAT1, CYR61 and MYC), promote EMT (ITGA3, AKT, LPAR3) or maintain a stable tumor microenvironment (IL-6, Csf1-3, CCL2, CSCL1/2). Distinct upstream regulators including MAPK signaling pathway and WNT5A regulate the nuclear translocation of YAP in either Hippo-dependent or Hippo-independent pathways.
