ecDNAs replicate along with chromosomes in the S phase, appearing as double minutes. During mitosis, acentric ecDNAs attach to chromosomes and segregate randomly, resulting in unequal ecDNA copies in daughter cells. Multiple rounds of cell division increase ecDNA copy number and heterogeneity.
