aPC-preincubation of mouse pan T-cells does not impair GvL effect. a, b Lethally irradiated (11 Gy)-recipient BALB/c mice were transplanted with 5 × 106 C57BL/6-derived BM cells, 5×103 GFP+MLL-AF9 (MA9) leukaemic cells, and 0.5 × 106 C57BL/6 T-cells without (B6T + BM + MA9) or with (B6T(aPC) + BM + MA9) aPC-preincubation. These groups were compared with the BALB/c mice receiving only allogenic BM and T-cells (B6T + BM) or receiving only allogenic BM and MA9 leukaemic cells (BM + MA9). In mice transplanted with aPC-preincubated T-cells and MA9 leukaemic cells (B6T(aPC) + BM + MA9) survival is improved (a Kaplan–Meier curve) and leukaemic load (determined by flow cytometry 1 week (b) and 4 weeks (c) after transplantation in peripheral blood samples) is markedly decreased; pooled data from three independent experiments each with four recipients.
