NPs suppress MHC class II expression for up to 6 weeks in transplanted human arteries in vivo. a Human arterial allografts pretreated with siRNA-NPs show reduction of HLA-DR on ECs for up to 6 weeks in vivo. Coronary arteries were incubated with siRNA-loaded NPs for 6 h and transplanted into SCID/beige mice. Three doses of hIFN-γ were administered every other day for 1 week. b After 1, 2, 4, or 6 weeks, arteries were harvested, sectioned, and stained using anti-CD3 antibody (red), anti HLA-DR antibody (green), and DAPI (blue), scale bar: 100 μm. c HLA-DR fluorescence was quantified from the images using ImageJ and normalized to CTL NP treated arteries.
