Proposed model of simple sugar preference regulation by neuronal SIRT1. Ingestion of simple sugar promotes secretion of FGF21 from the liver. The hepatokine FGF21 acts as an endocrine signal to Oxt neurons and promotes Oxt transcription (AKT signalling) and neuronal activation (ERK signalling). Oxt suppresses simple sugar preference by acting on OXTR. SIRT1 promotes FGF21 sensitivity in Oxt neurons and stimulates Oxt transcription through NRF2, thereby potentiating the negative feedback regulation of simple sugar preference through FGF21 and Oxt
