Fig. 6: Mathematical modeling shows native CRAC channels are ORAI heteromultimers.
From: The native ORAI channel trio underlies the diversity of Ca2+ signaling events

Model simulations used the parameter p (with units of µM) as the equivalent to increasing agonist concentrations. Other parameter values are I1 = 1, I2 = 0.2, I3 = 0.2 (all in units of concentration), K11 = 14, K22 = 1.5, K33 = 0.93, K12 = 19, K13 = 7, K23 = 14 (all in units of 1/concentration), and α11 = 2, α22 = 1, α33 = 0.7, α12 = 0.1, α13 = 0.1, α23 = 0.1 (all in units of 1/time). All other parameter values are as in ref. 22. a–c Model the response to increasing agonist concentrations in parental HEK293 cells and 5 different single, double, and triple ORAI-knockout cell lines. d Models the cytosolic Ca2+ response to maximal store depletion with thapsigargin in the absence then presence of 2 mM external Ca2+.
