HD-MSCs (MSC; blue) were co-cultured with MM.1S (orange) or RPMI-8226 (red) cell lines separated by a transwell system in the presence (darker shade) or absence (lighter shade) of CM-272. A Relative expression and B DNA methylation of genes RUNX2, SPP1, IBSP, and HOXB7 was assessed. Gene expression was by normalization against RPL38. Bar plots represent mean ± SEM of 3–5 independent experiments and statistical significance was calculated by paired one-tailed Student’s t-test (*p < 0.05, **p < 0.01). C Mineralization was assessed by alizarin red staining in differentiated OBs from HD-MSCs co-cultured with MM.1S or RPMI-8226 cell lines in the presence or absence of CM-272. Representative micrographs show matrix mineralization by alizarin red staining of corresponding differentiated OBs.
