a–c Capsule size is affected by the UGD1G19A mutation. Cells were stained by India ink and visualized by microscopy. The experiments were repeated 4 times independently with similar results. The capsule sizes of more than 65 cells were measured from 5 different fields for each indicated strain. Capsular thickness was defined as the distance between the visualized cell wall and the edge of the capsule. The box-and-whiskers plot is used to show median, quartiles (boxes), and range (whiskers). Two-tailed, unpaired t-test (****p < 0.0001). Bar = 5 μm. d UGD1 expression is downregulated in the Knrg1 containing strains. RNA was isolated and the expression levels of UGD1 were determined by qRT-PCR, normalized to the actin gene. The experiments were repeated three times. One-way ANOVA with Tukey’s test for multiple comparisons (ns = not significant and ****p < 0.0001).
