a, b Additional group I self-splicing introns were tested for expression of oGFP when introduced into oRiboT at sites 1 or 2. ce Fluorescence of oGFP-oRiboT variants with intron candidates at site 1 and Tetrahymena intron at site 2, 3, or 4. f RT-PCR was performed on total purified RNA from cells expressing oRiboT with Tfa, Tfb, or Tfc intron at site 1 or site 2 to determine whether introns were completely spliced out of oRiboT and ligated post-splicing.
