5-ALA and CAT were directly delivered to the tumor site by the transdermal co-delivery patch. The MN-CCPCA patch continuously catalyzed hydrogen peroxide (H2O2) into O2 to elevate the intratumoral sO2 level, which enhanced the physiological synthetic concentration and extended the METP of PpIX by “shutting down” PpIX outflow (downregulating HIF-1α and FECH) and upregulating PpIX biosynthesis (providing exogenous 5-ALA and upregulating ALAS). The platform of MN-CCPCA patch offers a strategy to monitor in vivo real-time PpIX and sO2 levels and optimize treatment parameters through duplex FL and PA imaging and ultimately improves therapeutic efficacy and biosafety of 5-ALA-PDT.
